Effect of estrogen signaling in CD8 T cell mediated antiviral response

چکیده

Abstract Estrogen, the major female sex hormone has been linked with immune response disparity between males and females. However, exact mechanism by which estrogen impacts our ability to fight viral infection development of immunologic memory remains unknown. CD8 T cells represent primary protective against viruses cancer. Here we aim understand how cell mediated antiviral is regulated estrogen. Systemic LCMV in ovariectomized mice generated lower virus-specific compared sham surgery controls. To check if this reduction intrinsic, I a transgenic receptor-α deficient mouse line (ESR1 KO). Using co-adoptive transfer model, differentiation ESR1 KO control both systemic local influenza infection. The lack signaling resulted reduced abundance effector context infections. also displayed cytotoxic ability. In vitrostudies revealed that expansion was primarily impacted signaling, whereas activation cellular apoptosis remain unchanged. Altogether, suggests crucial role steroid initial differentiation, significant ramification for generation responses pathological or physiological deficiency. HDAC1 Controls CD8+ T Cell Homeostasis and Antiviral Response

Reversible lysine acetylation plays an important role in the regulation of T cell responses. HDAC1 has been shown to control peripheral T helper cells, however the role of HDAC1 in CD8+ T cell function remains elusive. By using conditional gene targeting approaches, we show that LckCre-mediated deletion of HDAC1 led to reduced numbers of thymocytes as well as peripheral T cells, and to an incre...

-Terminal Kinase (JNK)1 and JNK2 Signaling Pathways Have Divergent Roles in CD8 T Cell–mediated Antiviral Immunity

c-Jun NH 2 -terminal kinases (JNK) play important roles in T helper cell (Th) proliferation, differentiation, and maintenance of Th1/Th2 polarization. To determine whether JNKs are involved in antiviral T cell immunity, and whether JNK1 and JNK2 bear biological differences, we investigated the immune responses of JNK1-deficient and JNK2-deficient mice to lymphocytic choriomeningitis virus (LCMV...

The Role of β7 Integrins in CD8 T Cell Trafficking During an Antiviral Immune Response

The requirement of beta7 integrins for lymphocyte migration was examined during an ongoing immune response in vivo. Transgenic mice (OT-I) expressing an ovalbumin-specific major histocompatibility complex class I-restricted T cell receptor for antigen were rendered deficient in expression of all beta7 integrins or only the alphaEbeta7 integrin. To quantitate the relative use of beta7 integrins ...

Identification and Visualization of CD8+ T Cell Mediated IFN-γ Signaling in Target Cells during an Antiviral Immune Response in the Brain

CD8(+) T cells infiltrate the brain during an anti-viral immune response. Within the brain CD8(+) T cells recognize cells expressing target antigens, become activated, and secrete IFNγ. However, there are no methods to recognize individual cells that respond to IFNγ. Using a model that studies the effects of the systemic anti-adenoviral immune response upon brain cells infected with an adenovir...

Antiviral Cd8+ T Cell Responses in Neonatal Mice

Polyoma virus is a potent oncogenic pathogen when inoculated into newborn mice of particular H-2(k) strains. Using D(k) tetramers containing the dominant antipolyoma CD8(+) T cell epitope, middle T protein (MT)389-397, and intracellular interferon gamma staining, we enumerated MT389-specific CD8(+) T cells in infected neonates having opposite susceptibilities to polyoma virus-induced tumors. In...
